Title: A next generation mathematical model for the in vitro to clinical translation of T-cell engagers
Abstract:
- Regulatory agencies recommend more quantitative justifications for clinical dose selection, especially for bispecific T-cell engagers (TCE) which form a “trimer” and which serves as a proxy for TCE activity.
- We present a next-generation mathematical model for TCEs where the key step in trimer formation is sensitive to the cell-surface density of TCE targets, rather than receptor concentration. This model is robust to common variances between experimental and clinical conditions which commonly make traditional models less accurate.
